Abstract

Escherichia coli is a Gram-negative bacteria and the main cause of diarrhea. One way to identify E. coli bacteria is by using safranin. However, safranin has several limitations, such as expensive and carcinogenic. So, alternative dyes based on natural ingredients are needed, one of which is the rosella flower (Hibiscus sabdariffa L). This research aims to determine the potential of rosella flower extract (Hibiscus sabdariffa L) as an alternative dye to replace safranin. The method used in this research was an experiment using rosella flower extract with concentrations of 50%, 60% and 70%. The sampling technique used was purposive sampling with a total sample of 27 samples. Statistical test results using the Mann Whitnay U test at a comparison of 50% and 60% showed that there was no difference in coloring quality with not good category (p value > 0.05), whereas at a concentration of 70% compared with a concentration of 50% and 60% it was found There are differences in the results with poor quality with a p value <0.05. Furthermore, the significance value between variations of 50%, 60% and 70% compared with the safranin control, showed that there is a difference in coloring quality (p value <0.05). This showed that rosella flowers have potential as an alternative dye to replace safranin but have not shown better results compared to safranin.
