Abstract
Triglyceride and cholesterol testing is a crucial component of lipid profile evaluation for diagnosing cardiovascular disease. Laboratory results can be influenced by pre-analytical factors, such as sample processing methods. This study employed a quasi-experimental design with a paired-sample approach, subjecting each blood sample to two treatments: allowing it to clot for 30 minutes prior to centrifugation versus immediate centrifugation. The study involved eight participants who met the inclusion and exclusion criteria. Triglyceride levels were measured using the GPO-PAP method, while cholesterol levels were determined using the CHOD-PAP method on a Dialab Clinical Chemistry Analyzer. Data analysis was performed using the Paired Sample t-test for triglycerides and the Wilcoxon test for cholesterol. The results showed that mean triglyceride levels were 115.538 mg/dL for samples allowed to clot before centrifugation and 120.087 mg/dL for those centrifuged immediately. The Paired Sample t-test yielded a p-value of 0.010 (p < 0.05), indicating a significant difference. Meanwhile, the median cholesterol levels were 152.00 mg/dL for samples allowed to clot before centrifugation and 160.00 mg/dL for those centrifuged immediately; the Wilcoxon test yielded a p-value of 0.206 (p > 0.05), indicating no significant difference. In conclusion, allowing blood to clot before centrifugation affects serum triglyceride levels but does not significantly affect serum cholesterol levels.
